The Effects of Cytokines on IL-2 Induced NK Cell and LAK Cell Activity



Abstract
Immunotherapy using IL-2 and IL-2 induced LAK cells have induced some tumors to regress in patients with advanced cancers. But life threatening toxicities of IL-2 in dose escalation was themajor obstacle in clinical application because high dose
of
IL-2
is required to get effective anti-cancer activities. Varieties of cytokines including ¥áinterferon, ¥ã-interferon, Il-4 and even cyclophosphamide have been used in combination with IL-2 in an attempt to augment cytolytic activity of LAK cells and
to
reduce IL-2 dose with effective cytolytic activity, and with little harmful side reactions.
GM-CSF, ¥á-interferon and ¥ã-Interferon were added to a culture of human peripheral blood mononuclear cells in the absence and in he presence of IL-2 to elucidate their effects on the induction of LAK cells and the cytotoxicity of LAK cells in
vitro. NK
cell sensitive K-562 cells and NK cell-resistant Daudi cells were used to measure for NK cell and LAK cell activity using 4 hr-51Cr release assay from tumor cells, respectively.
For LAK cell induction, IL-2 was most effective one and then ¥á-interferon the next. But ¥ã-interferon and GM-CSF have little effect on it. For the augmentation of NK activity, IL-2 was also the best and ¥á-interferon, GM-CSF and ¥ã-interferon in
decreasing order. The combination of IL-2 with ¥á-interferon and IL-2 with GM-CSF augumented the cytotoxicity of LAK cells. Like ¥á-interferon which has been previously known, the substantial cytotoxic activity.
In this study auther conclude that the combination therapy with IL-2/GM-¥ãCSF or IL-2/¥á-interferon as an adoptive immunotherapy would increase antitumor cytotoxic potentials and decrease IL-induced serious side-reactions.
